چکیده

Recent experiments on the light receptor bacteriorhodopsin have revealed the protein conductive properties and connected them to its sensing action. In particular it was shown that the super-Ohmic I-V characteristic acquired in dark, changes in the presence of green light, with an enhancement of current at increasing bias values. Here we propose a current transport model for proteins able to reproduce experimental data, mainly the dependence of current on their three dimensional (tertiary) structure. The model is based on a resistance network model and implements a tunneling mechanism of charge transfer between the aminoacids constituting the protein.

Charge transport in bacteriorhodopsin monolayers: The contribution of conformational change to current-voltage characteristics

When moving from native to light activated bacteriorhodopsin, modification of charge transport consisting of an increase of conductance is correlated to the protein conformational change. A theoretical model based on a map of the protein tertiary structure into a resistor network is implemented to account for a sequential tunneling mechanism of charge transfer through neighbouring amino acids. ...

Protein Conformational Changes in the Bacteriorhodopsin Photocycle: Comparison of Findings from Electron and X-Ray Crystallographic Analyses

Light-driven conformational changes in the membrane protein bacteriorhodopsin have been studied extensively using X-ray and electron crystallography, resulting in the deposition of >30 sets of coordinates describing structural changes at various stages of proton transport. Using projection difference Fourier maps, we show that coordinates reported by different groups for the same photocycle int...

Conformational flexibility of membrane proteins in electric fields. I. Ultraviolet absorbance and light scattering of bacteriorhodopsin in purple membranes.

Bacteriorhodopsin of halobacterial purple membranes exhibits conformational flexibility in high electric field pulses (1-30 x 10(5) V m(-1), 1-100 micros). High-field electric dichroism data of purple membrane suspensions indicate two kinetically different structural transitions within the protein; involving a rapid (approximately 1 micros) concerted change in the orientation of both retinal an...

Local-global conformational coupling in a heptahelical membrane protein: transport mechanism from crystal structures of the nine states in the bacteriorhodopsin photocycle.

Proton pumps utilize a chemical or photochemical reaction to create pH and electrical gradients between the interior and the exterior of cells and organelles that energize ATP synthesis and the accumulation and extrusion of solutes and ions. G-protein coupled receptors bind agonists and assume signaling states that communicate with the coupled transducers. How these two kinds of proteins conver...

Conformational change of bacteriorhodopsin quantitatively monitored by microcantilever sensors.

Bacteriorhodopsin proteoliposomes were used as a model system to explore the applicability of micromechanical cantilever arrays to detect conformational changes in membrane protein patches. The three main results of our study concern: 1), reliable functionalization of micromechanical cantilever arrays with proteoliposomes using ink jet spotting; 2), successful detection of the prosthetic retina...
